Since language-based diagnostic procedures may be confounded by cognitive-linguistic compensation strategies, nonverbal test materials were created, including human affective and vegetative sounds. Depending on video context, each sound could be interpreted either as direct expression of an agent’s affective/vegetative state or as result of intentional-executive mental operations. “Situational relevance” and “intentionality” ratings by a group of twelve healthy subjects nicely differentiated between context types. By contrast, an AS subject showed a systematic overinterpretation of vegetative/affective signals in terms of planned activities. Such overestimation of intentional motivation, leading to impaired social cognition, might be due to the inability to utilize “affective resonance” mechanisms for the interpretation of an individual’s internal state. The Autism Spectrum Quotient (AQ) and the Empathy Quotient (EQ) were used to measure autistic traits. The severity of depression was measured by Beck’s Depression Inventory. Results The group of chronically depressed patients showed significantly elevated autistic traits according to both AQ and EQ measures. In addition, 48.4% of the patients with chronic depression showed AQ scores within the range of the broader autistic phenotype. Similar scores were found among 3.2% of the healthy controls and 100% of the patients with autism spectrum disorder. Conclusions About half of the chronically depressed patients showed elevated autistic or autism-like traits. It remained unclear whether this was due to the state of chronic depression or a kind of premorbid autistic personality trait. The findings illustrate the need for further research to clarify the possible role of autistic traits in the development of chronic depression. Furthermore, they reveal that it might be clinically useful to focus on autism-like social impairments in therapy for chronic depression. Previous studies suggest an important role of these systems in the pathophysiology of ASD, including a study of our group reporting decreased glutamate concentrations in the pregenual anterior cingulate cortex (ACC) of adults with ASD. The aim of this study was to replicate our previous findings of impaired glutamate metabolism in ASD in a new sample and to additionally quantify GABA in the ACC and dorsolateral prefrontal cortex (dlPFC). Concentrations of GABA and glutamate-glutamine (Glx; combined glutamate and glutamine signal) were quantified in the ACC and dlPFC of 43 adults with ASD and 43 neurotypical controls (NTC) by magnetic resonance spectroscopy (MRS). The ASD group showed increased absolute GABA concentrations and elevated GABA/creatine ratios in the left dlPFC compared to NTC, while no group differences were detected in the pregenual and dorsal ACC. Previous findings of altered Glx concentration in the pregenual ACC of the ASD group could not be replicated. Regarding Glx concentrations and Glx/creatine ratios, there were no significant differences in the dlPFC and ACC either. The study supports the hypothesis of an altered GABA and glutamate equilibrium, indicating an imbalance between excitatory and inhibitory metabolism in ASD patients. However, inconsistent results across studies and brain regions suggest a complex underlying phenomenon. LAY SUMMARY: Adults of the autism spectrum exhibit elevated levels of the inhibitory neurotransmitter GABA in the left dorsolateral prefrontal cortex. This finding supports the hypothesis of an imbalance between excitatory and inhibitory equilibrium in patients with autism spectrum disorders.
